The parts below are derived from the ilux operon according to Gregor et al.’s study in 2018, which comes from the lux operon. At the beginning of the project, we already had the ilux operon, in a pGEX plasmid.
Our parts were obtained from diverse methods: fiatluxCD and the promoter BBa_J23117 were synthesized, whereas fiatluxABE was obtained by an overlap PCR (for silent mutations). The composite part fiatluxCDABE is made of fiatluxC, fiatluxD, fiatluxA, fiatluxB and fiatluxE.
You can find below a table with the different parts of the project FIAT LUX, created by our team INSA_Lyon1. If you need more details and precisions about our parts, you can contact us at igem.insalyon2022@gmail.com.

BBa_K4239001 | fiatluxC | Basic | fiatluxC is to be used with fiatluxD (BBa_K239002) and fiatluxE (BBa_K239005). It codes for a subpart of a fatty acid reductase. With the subparts coding for fiatluxD and fiatluxE, they form a complex that recycles fatty acids to fatty aldehydes. | 1443 bp | Works | 10-12-21-23-25 |
BBa_K4239002 | fiatluxD | Basic | fiatluxD is to be used with fiatluxC (BBa_K239001) and fiatluxE (BBa_K239005). It codes for a subpart of a fatty acid reductase. With the subparts coding for fiatluxC and fiatluxE, they form a complex that recycles fatty acids to fatty aldehydes. | 924 bp | Works | 10-12-21-23-25-1000 |
BBa_K4239003 | fiatluxA | Basic | fiatluxA is to be used with fiatluxB (BBa_K239004). It codes for a subpart of the luciferase protein. With the subpart coding for fiatluxB, they form the luciferase protein. | 1083 bp | Works | 10-12-21-23 |
BBa_K4239004 | fiatluxB | Basic | fiatluxB is to be used with fiatluxA (BBa_K239003). It codes for a subpart of the luciferase protein. With the subpart coding for fiatluxA, they form the luciferase protein. | 984 bp | Works | 10-12-21-23-25-1000 |
BBa_K4239005 | fiatluxE | Basic | fiatluxE is to be used with fiatluxC (BBa_K239001) and fiatluxD (BBa_K239002). It codes for a fatty acid reductase subpart. | 1113 bp | Works | 10-12-23-25-1000 |
BBa_K4239006 | fiatluxCD | Composite | Contains fiatluxC and fiatluxD together with the J23117 promoter. With the subpart coding for fiatluxE (BBa_K4239005), they form a complex that recycles fatty acids to fatty aldehydes (fatty acid reductase). | 2434 bp | Works | 10-23-25 |
BBa_K4239007 | fiatluxABE | Composite | fiatluxA (BBa_K239003) and fiatluxB (BBa_K239004) are to be used together. They code for the luciferase protein. | 3378 bp | Works | 10-12-23 |
BBa_K4239008 | fiatluxCDABE | Composite | The complete fiatlux operon is composed of 6 genes: fiatluxA (BBa_K239003), fiatluxB (BBa_K239004), fiatluxC (BBa_K239001), fiatluxD (BBa_K239002) and fiatluxE(BBa_K239005), that form an autonomous system producing bioluminescence thanks to the encoded enzymes. | 5867 bp | Works | 10-23 |
BBa_K4239009 | hok/sok | Basic | Hok/sok is a toxin/antitoxin system made to be used on the same plasmid than an interest gene (the fiatlux operon, our study). This method of selection ensures that all the bacteria present on the plant have the plasmid, and can hence emit light. | 427 bp | Works | 10-12-21-23-25-1000 |
BBa_K4239010 | axe/txe | Basic | Axe/txe is a toxin/antitoxin system made to be used on the same plasmid than an interest gene (the fiatlux operon, our study).
