Spider silk is formed from spideron with a suitable pH buffer and concentration of spideron.

After transformation, the transformed E. coli will be cultured on a large scale in a large shaking incubator/fermenter. And then, doing protein expression, protein extraction and purification, spider silk can be formed by squeezing the syringe which contains spidroin. By repeating the above steps, a large amount of spider silk is collected. Next, we would test the properties of spider silk to study their potential application, like strings. And then, by mixing different spidroins or proportional expressions, the properties of spider silk can be modified. Moreover, a SpyTag gene was added in one of the gene, NT-2Rep-CT, to increase the feasibility of forming different properties of silk.
