All of the data collected above has been planned in advance and with the consent of the PI to determine that there are no ethical issues at the event. Additionally, all data and photos, videos, etc. have obtained the consent of the person concerned.
Liquid nitrogen
In order to preserve the sample better, we need liquid nitrogen to cool the material in the experiment. In order to ensure the safety of the experimenter, we need to understand some precautions when using liquid nitrogen. Liquid nitrogen in our laboratory is provided by a regular company. In the process of liquid nitrogen dumping, two or more laboratory personnel are generally required to complete it together. If liquid nitrogen is used for the first time, it is generally completed under the guidance of experienced personnel. At the same time, to avoid frostbite caused by direct contact of liquefied gas with skin, eyes, hands and feet, the laboratory personnel will take necessary protective measures, wear protective masks, protective glasses, and leather gloves and other protective equipment. When the sample is removed from the liquid nitrogen, the experimenter will wear cotton gloves and use long tweezers to pick the sample out and place it in the box. If liquid nitrogen is accidentally applied to the skin, soak the frostbitten part in warm water not more than 40℃ for 20 ~ 30 minutes, and then elevate the frostbitten part. At room temperature, do not bandage anything and keep quiet. Severe frostbite should be treated by a professional doctor
Training
1.On-line Our members have systematically studied laboratory safety in the safety education
for college students on the Learning platform.
2.Certificate Our members took the laboratory safety examination of agricultural Life
Science on November 16, 2021, and the certificate of qualification will be issued if the
score is above 95. Only those who hold the certificate can enter the laboratory.
3.Offline Our members have operated the machine under the leadership of doctoral students
and graduate students.
Epidemic
Since the outbreak of COVID-19 in 2020, the epidemic is still not over. In response to the national epidemic prevention policy, our team members wear masks in and out of the laboratory, maintain appropriate social distancing, frequently wash their hands with alcohol-based hand sanitizer, receive comprehensive vaccination, and take nucleic acid tests regularly to ensure our personal safety. Luckily, we didn't get infected during the game.
CRISPR-Cas9 gene editing
We genetically modified Longan callus using CRISPR-Cas9 genome editing technology, which is different from the traditional genetic modification technology. Unlike genetic modification, gene editing only changes parts of a crop's original genes, without introducing foreign genes. Because of this technology, officials say there is no problem with safety and that no harmful substances will be produced.
Nucleic acid
In molecular biology, an effective method for nucleic acid separation, purification and protein recognition in electrophoresis experiments can be used in conjunction with gel imaging systems or alone, depending on the purpose of the experiment. AGAR gel electrophoresis is an experimental method to judge the molecular weight of DNA by the electrophoresis rate of DNA fragments in the electric field of agarose medium. Nucleic acid dyes are essential in the preparation of agarose. However, nucleic acid dyes used in the laboratory are toxic. In order to minimize the toxicity of nucleic acid dyes to the experimenter, the experimenter should wear masks and gloves throughout the operation, and change the gloves in time.
Agrobacteria
Tumefaciens tumefaciens and Tumefaciens tumefaciens inducing cells contain Ti plasmid and Ri plasmid, respectively, on which there is a T-DNA(transferring-DNA) transferring-DNA, which can be inserted into the plant genome after Tumefaciens enters the cell through the wound of the plant. And can be stably passed on to offspring through meiosis. In the experiment, Agrobacterium tumefaciens was used. In order not to cause biological contamination to the environment, the used materials were autoclaved and processed in a centralized manner in strict accordance with laboratory regulations.

Add the following ingredients to the beaker and stir with a glass bar.
CMC 0.12g
White granulated sugar to be determined
Pectin 0.12g
When the solution in the pot is about to boil, reduce the heat, pour the solution in the
beaker into the pot, stir while adding, and then boil for 2-3 minutes. After cooling,
determine whether the sweetness and acidity are appropriate, to determine the amount of
citric acid and granulated sugar.
After determining the amount of citric acid and white granulated sugar, repeat the above
operation, and then finally boil and turn off the fire, add the right amount of essence, and
taste after cooling, to determine whether the fragrance is appropriate, so as to determine
the amount of essence.

Pre-experimental results: after repeated tasting by the experimental
members, it was
determined that the amount of citric acid is about 0.11%, the amount of white granulated
sugar is about 1%, and the amount of essence is about 0.0002% to drink the best taste.
Several controlled clinical trials were conducted in the beverage
processing laboratory.
After several adjustments, we have strictly controlled the content of additives and
determined the range of additives that can roughly comply with food safety laws.
