Overview

At present, the diagnosis of intestinal microbleeds is mainly fecal occult blood test and colonoscopy.

Colonoscopy is not only expensive, but also can cause psychological pressure on patients, making people feel physically uncomfortable, and also has certain safety risks. Once intestinal perforation occurs, the damage to the body is very huge.

Although the cost of fecal occult blood test is low, it requires patients to prohibit eating meat, food containing animal blood and certain vegetables or taking iron and vitamin C three days before the test. Not only the error is large, but also there are many influencing factors. It is likely to have false positive results, and the accuracy of the detection is low, which is not conducive to follow-up treatment.

Fecal occult blood test kit

In China, many poor areas have poor medical facilities and the traffic there is also not very convenient, so it is difficult to go to regular hospitals for the appropriate inspection. Therefore, convenient detection device is particularly important. Based on cost and accuracy considerations, this year NWU-CHINA-A designed a simple isolation and culture device that can detect intestinal microbleeds under non-experimental conditions.

Simple hardware device design under non-experimental conditions

The following is the detailed description of the hardware.

1.We placed a filter membrane with a pore size of 5μ in the front of the syringe, loaded the stool sample we needed to detect into the syringe, and squeezed it into the culture device.

2.In the culture section, we used a transparent cylindrical plastic box, in which to add the culturing medium. We used liquid and solid media. The following is the ingredients of the medium.

①Liquid ( 20ml ) : dry powder 0.5g,Tryptone 0.2g, Yeast extract 0.1g, Sodium chloride 0.2g, antibiotic K50 2μl, water 20ml.

②Solid ( 20ml ) : dry powder 0.8g, Tryptone 0.2 g, Yeast extract 0.1 g, Sodium chloride 0.2 g, Agar powder 0.3 g, antibiotic K50 2 μl. If we want to use it, we need to add 20ml of water and turn it into a liquid.

3.We purchased a heating belt with vibration and heat preservation wrapped on the outside of the plastic box for heat preservation and oscillation to promote the growth of the strain.

The above is a simple hardware device under non-experimental conditions. In our communication with DKU_China, we all wanted to design common hardware that matched both team projects. This hardware was finally designed and printed by DKU_China.

Here are the specific instructions for the use of this biosafety box.

1. The small hole of the safety box is 6mm in diameter and the big hole is 10mm in diameter. The volume of each small hole was 1.131ml.

2. After sampling, the fecal samples are placed into the reagent tubes in the large wells. And the configured 1ml of culture medium liquid is placed directly into the small wells before using.

3. In the specific experiment, the stool samples in the large hole are quantitatively mixed into the small hole with a pipette. The safety box is put into the incubator and incubated at the appropriate temperature. Finally, judge whether there is intestinal bleeding by observing whether there is a color change.

4.Use screws to closely bind the lid and the box to prevent bacteria from overflow when shaking.

So far, we had built a simple device and tested it. Our design was from the perspective of patients and we strived to improve the application value of the product. Our products are not only convenient to use, but also low in cost. It can be used in families, small clinics and other occasions to help patients detect intestinal microbleeds as early as possible and treat them in time. In the future, we will continue to optimize our products to make their use more convenient and accurate.
