Proof of concept


The deletion of perR improves the oxygen tolerance of Clostridium tyrobutyricum.

This year, NJTech_China has realized the aerobic growth of Clostridium tyrobutyricum. To prove the concept, we set a series of experiments to prove the experiment is success.

Sequencing verification

We did sequencing verification of obtained mutant. The results showed that the perR sequence was deleted in Ct (ΔperR), compared with Ct (Control).


The growth profiles

We tested the growth concentration of Ct (ΔperR) and Ct (Control) under different contents of oxygen and drew the growth curve. The profile shows that Ct(ΔperR) has a significant improve comparing with Ct (Control). Moreover, the higher the oxygen concentration, the greater the growth performance of Ct(ΔperR).
Physiological characteristics

We measured the ROS levels in the logarithmic prophase. From fig A, the intracellular ROS levels in Ct (ΔperR) is lower than Ct (Control) in the presence of oxygen. Also, the ratio of NADH/ NAD+ in Ct (ΔperR) is lower than Ct (Control) in the presence of oxygen respectively, during the early-, mid- and late- logarithmic phase, from fig B.

The survival rate of Ct (ΔperR) has significant improve than Ct (Control) under oxidative stress.
