Safety

Safety is a key aspect of our project. Given that our goal is to design and develop a smart home system based on an E. coli light control system, which aims to regulate user’s mood and relieve fatigue. On this page, we will show how we review and subsequently implement different measures to ensure security for all participants from development to use.

1. Lab safety


All laboratories in China must be certified by the China National Accreditation Service for Conformity Assessment. This year, the project was conducted at the BSL-1 lab, where we met all the fire and safe production requirements. In addition, members of the research team wore lab suits, gloves and goggles during the experiment. Bacterial work was performed only in biosafety cabinets.


1.Microorganisms review


The E. coli strains we used: DH5a, BL21 and Top10, which do not have serious diseases or other problems to humans or the environment, belong to BSL-1 organisms.



2.Experiments


Our E. coli. will be designed to interact with light. They contain red and blue light promoters. Under the blue light irradiation, the promoter was acvited, and the genes such as KdcA and Adh1 were expressed, so that the target substances -- 2-PE was successfully expressed in E. coli. In addition, simultaneous irradiation of blue light and red light will trigger toxic proteins and kill bacteria.


When cooperating with genetically modified organisms, the engineering bacteria involved in the project will be strictly isolated from the external laboratory environment. After the experiment, self pressure steam sterilization and dry heat sterilization will be used for sterilization, and then placed in the designated experimental waste collection point to avoid environmental pollution by genetically modified organisms. In addition, all biological pollutants are submitted to autoclaving and treatment.



The reason why we can complete all the experimental tasks is due to Jilin University, which provides us with a biosafety level 1 laboratory. There is a biosafety cabinet in the laboratory, which allows us to work in sterile conditions. We have received guidance on general laboratory safety and how to deal with hazardous situations related to laboratory practice, chemical, electrical and fire safety. Another important point is the correct management of waste and different containers.


3.Harmful reagents and procedures


Some planned experiments must be carried out using harmful reagents or procedures. Team members must strictly abide by the biosafety law of the people's Republic of China and the laboratory safety management measures of Jilin University to prevent harm to themselves or the environment taught in the safety training.

Use ultraviolet rays to disinfect the biosafety cabinet and the equipment in it. All safety measures were taken to avoid direct exposure to ultraviolet rays. For the disinfection and sterilization of hydrogels, ultraviolet disinfection and sterilization is used, and standard laboratory procedures are strictly followed.

Ethidium bromide will be used in the production process of electrophoresis gel. Since this chemical is a strong mutagen and has moderate toxicity after acute exposure, the team members have complied with all the safety requirements of the material safety data sheet (MSDS). In addition, we will use two layers of gloves to avoid skin contact with these chemicals.

Use IPTG, hydrochloric acid, mercaptoethanol, acrylamide, 95% ethanol and other chemicals, which are toxic. Take appropriate safety measures and dispose according to the disposal scheme.


4.Hardware Safety


Operators should receive safety education before using the equipment, and fully grasp the relevant operating procedures. Before the experimental equipment is connected to the power supply, the operator must carefully check whether the equipment condition and circuit connection condition are good and correct before switching on the power supply and starting the equipment. When operating, the operator must wear insulating gloves, work shoes, and some necessary protective tools, and under the guidance of qualified professionals, in accordance with the correct operating procedures to prevent electric shock accident. After the completion of the operation to do a good job of site cleaning, cut off the power of all equipment before leaving.

2. Data security


During the implementation of human practical activities, we received human related data. We follow the general data protection regulations, and the measures we take in specific cases will be further explained below.

This project does not collect the user's name, location, email or any other information that can be used to identify someone. The data is only collected by the camera. The collected data includes micro expressions, heart rate, etc. this information is only used to characterize the sample. Since these matters are sensitive information, the information is handled with the highest security. Before the program runs, the user will be prompted to provide camera permission. The user's personal information will only be saved locally and deleted after the program runs.


3. User Safety


After preliminary detection of gas components, we found that no other gas was detected except 2-PE. 2-PE is widely used in food and perfume. Our synthetic amount is within the safe range and harmless to human body. In toxicity tests in rats and rabbits, a liquid dose of more than 5000 mg / kg will be fatal. The amount of gas released from our experiment is low, which can regulate mood and has no other toxic effects.

Considering that the release of chemical substances may be suspected of abusing chemicals or have some indirect effects on human body, we choose some extracts, which are essentially aromatic compounds or mixtures extracted from plants, rather than substances made in direct industry such as nicotine, which can eliminate this effect to a certain extent.


4. Environment Safety


We synthesized the plasmid and transferred it to E. coli. The treatment of synthetic strains may cause environmental pollution, but we will avoid this situation by taking reasonable steps and optimizing the treatment process. ​

We need to mail samples between two laboratories in different cities. We will properly package the samples to prevent the bacteria resistant to chloramphenicol from leaking from the packaging.
