Based on the Anderson Promoter Library and the Rackham RBS library, we believed that using the understanding of the 5'UTR and it's working we could synthesize a library of it that gives us a range of expression values. This library can be used and taken forward by anyone. You need to use BBrickIt v.2 developed by us to BioBrick your desired 5'UTR from our collection according to RFC[10] standards. The BBrickIt v.2 is open-source and available on our software page. You can use online tools to make your reverse oligo and forward strands. These can be ordered from companies like IDT. Once you have your oligos, you need to construct a double stranded 5'UTR. This is available in our Protocol attached on the Wiki. We are further pursuing this and taking it forward to the Biotechnology industry. The variations received in the expression values ratifies is in sync with our experimental design and expectations giving a valid proof of concept. This is our contribution to the iGEM Registry.
