Sustainability is at the core of our team.In the face of rapid global transmission of multi-resistant and pan-resistant bacteria (also known as "super-bacteria"),this phenomenon has become one of the important threats to hinder global sustainable development.Our central goal is to solve the current deficiencies in nucleic acid detection by developing rapid, high sensitivity and strong specificity detection methods, and to improve the specificity detection of CRISPR / Cas (Cas13 and Cas14) nucleic acid detection methods. This technology has the potential to be widely used in Marine aquaculture, medical and health care, food safety, disease prevention and control.According to 17 Sustainable Development Goals (SDGs),we have identified four of them that corresponded the most to our problems and have tried to comply with them as much as possible. These are:

Goals 2 Zero Hunger

Detection of pathogenic microorganisms, such as hepatitis A virus in shellfish, norovirus detection, detection of African swine fever virus in pork products and detection of Staphylococcus aureus in milk powder, etc. These tests play a very important role in daily food safety, and nucleic acid molecular detection methods are also used in more food hygiene testing with its accurate and rapid advantages. The portable and high-precision rapid detection will help to better achieve the goal of zero hunger sustainable development. According to incomplete statistics, aquatic diseases have led to losses of nearly 20 billion RMB in aquaculture in China, moreover affecting millions of practitioners (China Fisheries Yearbook 2019), of which fish account for about 55%-77%, crustaceans for 11%-28%, and shellfish for 3%-16%. After detailed research with project stakeholders aquaculture companies, we found that microbial resistance testing has become an area that companies must focus on and invest a lot of effort in. We also learned from the relevant person in charge of the Food Supervision Bureau that rapid testing is an effective measure for health protection and emergency response at large events: in the health protection of large events, in order to prevent the occurrence of mass food poisoning. Rapid testing is a necessity in China: China still has a long way to go in improving the overall level of food safety, and rapid testing will play an active and effective role in this.

SDGs 3: Ensuring a healthy lifestyle and promoting the well-being of people of all ages.

If effective measures are not taken, it is estimated that by 2050, 10 million people will die of drug-resistant bacteria worldwide every year, and the economic loss will reach 100 trillion US dollars. Microbial drug resistance has become a global public health crisis.

The immediate detection and identification of target pathogenic microorganisms is key to achieving disease control and treatment, and the development of rapid, sensitive, and accurate in situ assay technologies and platforms will improve the detection, identification, and risk control of pathogenic microorganisms. Currently, in situ assays still have many limitations, including sensitivity, cost, and the need for equipment and specialized personnel.

Our project achieves detection without PCR amplification by tandem association of Cas13a with Csm6 and parallel association with Cas14a, and accurate detection down to a single base by tandem association of Csm6, and then masks the interfering term using shackle PNA. It is less time consuming, less costly, more accurate and more sensitive than the fM detection limit of conventional PCR.

At the same time, we have developed a portable enzyme marker for this technology, which is lighter than the traditional enzyme marker, and does not need to be connected to a computer to use, and does not require pcr amplification, and designed a simple and friendly human-computer interface, so that people without basic biotechnology can quickly get started to use

Goals 4 Quality education

Team:Lambert GA/Sustainable - 2020.igem.org

Inspired by the Lambert GA team, we reached out to Hainan Foreign Language School, affiliated with Beijing Foreign Studies University, to conduct an online synthetic biology science lecture series - a knowledge series on crisper/ cas. Our team researched high school courses that could be integrated into our program, including biology, chemistry, and biotechnology at all levels. We spoke with several different teachers who showed interest in our project and mentioned that they would consider incorporating our project into their coursework. In the second year of the program, we plan to work with local high schools to build an out-of-classroom practice platform that will be used to stimulate further exploration of Crisper technology and provide new ideas for optimizing current instrumentation.(see education and communication)

The harm of marine drug-resistant bacteria outbreaks to marine resources SDGs

SDGs 14: Conservation and sustainable use of oceans and marine resources for sustainable development. In recent years, epidemic and outbreak of microbial diseases in mariculture have occurred worldwide, which has seriously restricted the healthy development of blue agriculture。Since Fleming's extraction of penicillin in 1929, research on antibiotics has advanced rapidly and is commonly used in the study and treatment of biological diseases.However, with the development of aquaculture and fisheries and other industries, the phenomenon of antibiotic abuse continues to emerge. According to statistics . In recent years the annual use of antibiotics in the world up to 100 ~ 200,000 tons.

The rich biological and mineral resources in the ocean have a great dependence on the marine environment in the long-term formation process. In general, the marine environment has its own balance, and when the marine environment is infringed by land pollution, this balance is broken, and the rich marine resources will be difficult to maintain the original existence of the situation, thus causing huge losses in the development of marine resources and seriously affecting the economic benefits brought by the development of marine resources.

Future Prospect

Sea Ranch-Ocean takes three-dimensional, recycling and ecological farming as the core, realizes "fish, shrimp, shellfish and algae" multi-nutrient level scientific multiplication, builds a marine breadbasket, and realizes sustainable and coordinated development. We cultivate algae farms and plant corals to increase marine biodiversity, repair marine ecology and form a virtuous cycle. Underwater set optical, acoustic intelligent marine microorganisms, composition detection equipment, real-time transmission detection data collection, the establishment of a good and stable underwater ecological cycle system.
