Overview:
Our project is inspired by COVID-19 and aims to apply it to the detection of drug resistance in marine microorganisms. We aim to build a single-base, amplification-free, portable nucleic acid detection platform, combined with CRISPR/Cas detection technology, so we pay close attention to safety throughout the development process.
Laboratory Safety
1.Experiment
The laboratory used for wet experiments is located in the State Key Laboratory of Marine Resources Utilization in the South China Sea of Hainan University. According to the requirements of the laboratory, all participants need to participate in the laboratory safety entrance examination before entering the laboratory. And our labs are supervised by a tutor system, and all experiments are conducted under the supervision and guidance of Wan Yi.
2.project
Although our project is to detect drug resistance of marine microorganisms, we strictly abide by the Convention to maintain biosafety and human health, so we have conducted virtual experiments in our laboratory with our own designed RNA and verified that it is not dangerous by evaluating various aspects.
3.Reagent materials
Toxic chemicals such as IPTG, β-mercaptoethanol, and polyacrylamide will be used in the process of this project moving forward, and we will take appropriate safety measures for them and dispose of them in accordance with the Hainan University Laboratory Safety Access Disposal Program.
4.Environmental safety
We obtain the target protein using E. coli BL21 (DE3), a synthetic strain that is non-toxic and engineered, and we also handle it properly during the experiment to ensure its closed-loop operation in the experiment and finally sterilize it.
Hardware Security
The hardware equipment of this project is mainly enzyme standard instrument, in hp's work and investigation, we finally choose the design material - polypropylene, which can make the instrument smaller and simpler in structure while making it safe. At the same time, it has good chemical resistance, heat resistance, electrical insulation, high strength mechanical properties and excellent wear processing properties, which provides good safety for our overall testing and subsequent corresponding processing.In addition, considering the diversity of user use, we have developed a set of corresponding post-use handling manuals, and through this strategy we can well avoid environmental pollution from the hardware.
