Safety
1. Masks and gloves must be worn at all times during the experiment.
2. Short skirts, shorts and slippers are not allowed.
3. Lab coat must be worn.
4. Laboratory coats are not allowed to be worn outside the laboratory or in public places.
5. Wash your hands after the experiment, whether you eat or not.
6. Eating/drinking is not allowed in laboratories where chemicals are used/stored.
7. Check whether the flask holder is firm before using the constant temperature culture shaker, pay attention to the symmetrical balance of the sample when using it, and the sample should not be full.
8. The sample must be mixed moderately before measuring by the ultra-micro nucleic acid quantifier. After each measurement, clean the upper and lower fiber ends with DEPC water, and then shut them down.
9. When using the electronic balance, it is necessary to put the weighing paper on first, subtract the weight of the tube, and then measure it. (Special attention: if there are drugs scattered in the electronic balance, it must be cleaned in time to prevent corrosion of the balance).
10. Take appropriate personal protection when using the biosafety cabinet. Wear personal protective clothing when using biosafety cabinets. Before and after use, need to shine ultraviolet light and wipe the table with alcohol.
11. After using the alcohol lamp, it must be put out with the lamp cap. Do not blow it with your mouth because it may explode. Also, do not knock over the alcohol lamp, in case spilled alcohol burns on the table, should immediately cover with a damp cloth.
12. When using the centrifuge, pay attention to the balance.
13. When using the pipetting gun to absorb liquid, it is necessary to loosen the thumb slowly and smoothly, and it is not allowed to loosen suddenly, in case the solution is sucked too fast and rushed into the liquid injector to corrode the plunger and cause air leakage.
14. Carefully check the previous use record before using the electric rotary instrument. If any abnormality is found, stop the test immediately and seek professional maintenance. Secondly, after use, do a good job of recovery and cleaning after use, and close the instrument.
15 Under normal circumstances, pH meter instrument in continuous use, to calibrate once a day; Generally, the instrument does not need to be calibrated within 24 hours. During the measurement, the leading wire of the electrode should be kept still, otherwise, the measurement will be unstable. Do not immerse the electrode in distilled water.
16. (important!!) When using the autoclave, confirm the correct connection of the POWER supply, turn ON the POWER switch ON the right side of the autoclave, and then press the "POWER ON/OFF" button to start the autoclave. When the pressure in the pot is 0, gently press down the handle of the sterilizer cover, step on the cover lock to release the pedal and open the cover. Fill the sterilizer pot with water (distilled water) until you see water over the bars of the center hole in the chassis. Place the item to be sterilized, close the cover and seal it with a magnetic seal, then lock the cover. Be careful not to overfill, do not have individual small particles.
17. The bacteria (Streptococcus Brevis, Escherichia coli) and plasmids exposed to during the experiment are harmless and cannot infect humans. We need to note that the reagents exposed during the extraction of plasmids may be harmful to the human body, so do a good job of protection.
