mApple is a luminescent protein, a variant of mOrange (E2050) discovered in 2008, that is substantially brighter than many other members of the RFP family. It is new to the iGEM registry and is the main reporter in our project.
A. caldus csoR is our best basic part because it was well documented with information regarding its characterization. It is a repressor sequence that functions in the copper buffer system of A. caldus. It was described and analyzed very recently, and we plan to explore its characteristics more.
PIII was identified as a possible promoter to pair with A. caldus csoR as a copper-sensitive operon. The resulting PIII-csoR system indeed induces strong transcription. However, it was reported to be non-copper-sensitive, a fact verified by our characterization of PIII. We therefore conducted mutation on the PIII-RBS system and constructed the K4494003-K4494098 mutant collection.

In order to improve an existing part to act as our benchmark, we characterized pCusC (K1980004). After the characterization procedure, we decided to further improve it. Our mutation generated 4 variants, three of which (K4494099-K4494101) do not function properly. However, K4494102 was a strong constant promoter, which could be seen as a successful improvement of K1980004.
This is the original sequence for our mutant collection K4494003-K4494098. It consists of PIII (K4494002) and an RBS (B0034) combined using Golden Gate Assembly. Since it has important significance in our project, we made it our Best Composite Part.
We requested this sequence from the authors of a paper on biosafety, but later abandoned the plan of using it due to changes in our project. However, we still thought that it might be useful to future iGEM teams who want to construct a biosafety circuit, so we kept the synthesized part to send to any future teams in need. This part is a member of the NWU (Northwest University) Part Library.
This is the transcriptional unit for A. caldus csoR that we used in our project. It works when combined with a corresponding promoter unit.
