Parts

    We use and construct parts that are considered an important part of our project, and most of them have been tested to ensure their reliability.
    To efficiently obtain these parts, we asked Tsingke Biotechnology to synthesize some of them. Besides, we also used colony PCR to get several parts from the original bacteria strain directly.

Basic Parts

    Among the basic parts designed this year by ourselves, BBa_K4144004 and BBa_K4144011 are our favorite. BBa_K4144004 is a codon-optimized eukaryotic protein, which transports SAMe from engineered bacterial cytosol to extracellular environment. BBa_K4144011, an optimized SAMe synthetase which will not be regulated by feedback inhibition, is utilized to improve Sam2 expression efficiency in gram-positive bacteria.

Composite Parts


    We have designed a series of composite parts this year and most of them have been tested. BBa_K4144081 and BBa_K4144082 are for the verification using EGFP of chemical switch and temperature switch respectively. BBa_K4144006 is for reducing the leakage of Pxyl and BBa_K4144008 is for synthesizing SAMe. BBa_K4144007 and BBa_K4144010 are for transporting SAMe. BBa_K3482025 is for oscillation of SAMe.BBa_K4144044 and BBa_K4144043 are for double selection and improved expression of LacI respectively. BBa_K143057 is used to express our SAMe transporter opPet8p.
