activities together.We were passionate about our projects though our communication was almost online due to the impact of the COVID-19.
In this process, we understood the importance of communication and mastered many effective

. In the whole process of cooperation and communicantion, we learned from each other and achieved each other's success.
We have maintained a close cooperative relationship with XJTLU-CHINA since the first communication on July 16, 2022. Our projects are all about
sewage treatment
. There are many aspects for us to learn from each other which both in experiments and hardware. At the same time, our team also sincerely thanks XJTLU-CHINA for their help.

XJTLU-CHINA has rich experience and has achieved great success, which has provided many guiding suggestions for our project.
On July 28, 2022, we went to Beitou Zhongmeng Water Purification Governance CO.LTD, Guangxi Nanning, China to
investigate

They helped to expand our

sample to investigate the water pollution and treatment in different areas.

which provides an exchange platform for 2022 freshwater restoration related iGEM projects and various teams jointly carry out some HP activities and education sections.

We fully understand each other's projects. XJTLU-CHINA uses

to adsorb heavy metal ions.

Our project of a
blue-green light
regulated biological bacteria preparation is an engineered bacteria transformed by means of synthetic biology.
We have rich experience in the
modification
of engineered bacteria. We shared our experience and put forward some guiding opinions to them.
3.Modeling
We shared our experience and ideas in the field of SimBiology.
We believed that the continuous communication between our two teams on modeling issues will make the modeling work of the two teams more and more smooth.
4.Safety
We shared our ideas with them,like suicide mechanism of our engineered bacteria and fully enclosed hardware space.
ZJU-China and our projects both use
Bacillus subtilis
as biological chassis. After the online meeting, ZJU-China got in touch with us and hoped to further cooperate in experiments. So we began to learn from each other and help each other in experiments in June.
Our communication and cooperation with ZJU-China mainly revolved around
experiments
. We learned from each other in the selection of
experimental materials
and the subsequent experimental
verification
. With their help, our project went more smoothly.
ZJU-China→GXU-China
1. We received their plasmid pHY300PLK-Pveg-GFP.
2. We successfully obtained the GFP gene we wanted from the plasmid pHY300PLK-Pveg-GFP in the later experiment.
GXU-China→ZJU-China
1. In June 2022, we sent the plasmid pHY-P43 to ZJU-China for their experiment.
2. We helped them to carry out the plasmid validation .
Water pollution is an extremely serious problem nowadays. Our iGEM teams also has many projects related to sewage treatment. We got in touch with them to exchange experience in the competition and discuss the experimental process.
We also set up
a Freshwater Bioremediation Alliance
for further exchange and cooperate. The teams participating in the alliance include Nanjing_NFLS, XJTLU-CHINA, Nanjing-China, CHINA-FAFU, GXU-China, UM_Macau and NPU-CHINA.
The aim of the alliance is to provide
a platform for communication
on iGEM 2022 freshwater restoration related projects and we can jointly develop some HP activities and educational sections.
On 27 July 2022, Nanjing_NFLS, XJTLU-CHINA, Nanjing-China, CHINA-FAFU, GXU-China and UM_Macau officially formed the Freshwater Bioremediation Alliance, which includes three main modules, science promotion, joint interviews and questionnaires.
At this meeting, we discussed the cooperation idea on the establishment of the alliance. Then, each team gave a brief introduction of their project, including experimental design, HP planning and hardware facilities, etc.
The aims of the meeting:
The main purpose of this meeting is to discuss our safety from three aspects.
1. We aim to prove that our experimental steps meet the safety standards.
2. We aim to prove that our engineering bacteria will not cause leakage.
3. We try to ensure that our projects will not cause harm to the environment and human beings.
Our proven of the safety:
1.The
Bacillus subtilis
used in the project will not cause harm to the environment and organisms.
2. We experimentally designed a light control switch, which activates the suicide mechanism when green light shines, and then kills bacteria.
3. The hardware adopts closed-loop design and the detection devices of temperature, pH, nitrogen oxide, etc. are designed, which ensures the safety to a great extent.
4. The harmful gases that may be discharged during the experimental detection process are collected and processed in a centralized manner so as to avoid being released to the environment.

We were honored to invite Professor Shen to give detailed answers to the questions raised by each team and also put forward some
practical suggestions
about our projects.

Our team asked some questions about the experiment and modeling. Without doubt, we got a lot of
feasible advice
from Professor Shen. We also
improved our project
with the help of the useful advice.
