February
- Our team was formally founded.
- We looked through the previous excellent projects and learned the basic notion of synthetic biology and iGEM. After comprehensive survey and discussion, we decided our project theme. We were then divided into three groups to implement our project separately. We held meetings and discussed every weekend about everyone’s progress and solved problems together.
3.19
- We celebrated the birthday of our three members together. Happy Birthday, Shuwan & Yue & Yiran!
- It’s the first birthday party of our team this year.
April-May
- We learned the fundamental knowledge of molecular cloning and laboratory qualities. After that, we were trained for molecular cloning operations, which helped us to design experiments rationally.
- We optimized and constructed the sequences of the needed genes.
- We completed the original hypothesis testing experiments needed, then consulted professors for advice. With their help, we gradually refined our project.
April-May
- Interview with Prof. Lina Wu
We interviewed Ms. Lina Wu who’s the associate professor of Xiamen University. She gave us a lot of advice on the design of OMVs.
- Fisheries Bureau
We contacted the Fisheries Technology Promotion Station of Fujian Provincial Fisheries Bureau, and conducted an online interview with Mr. Lin who is the deputy director of the laboratory.
May-June
BBa_K4195135, BBa_K4195136, BBa_K4195137 were constructed successfully.
6.15
- Our team was officially accepted by iGEM.
6.10-6.26
BBa_K4195116, BBa_K4195117 were constructed successfully.
6.27-7.8
BBa_K4195138 was constructed.
7.5
- Two first-year teams from Fujian Agriculture and Forestry University (FAFU) contacted us hoping for a visit. We accepted their invitation and held an offline seminar at Xiamen University. After the meeting, we also helped them contact professor Chenyun Guo for an interview.
7.7
- We are all sincerely believed in the power of prayer: everyday before doing experiment, we turn facing the South Putuo Temple, and wish today everything will go well.
7.16
- We were invited by GXU-China to take part in an online symposium held to share projects among a few teams. Grabbing the chance to discuss the prototype of project with experienced teams, we took part in the symposium.
7.18
- After induction, the mortality of engineered bacteria carrying kill-switch system: 0%
7.9-7.20
- BBa_K4195120, BBa_K4195121, BBa_K4195114, BBa_K4195115 were constructed successfully.
7.29
Give up purifying OMVs using nickel column. Thank you for nothing, high throughput Ni-based IMAC.
7.21-8.2
- BBa_K4195164, BBa_K4195165, BBa_K4195166 were constructed successfully.
Interviewing Prof. Shaowei Li
We visited Professor Shaowei Li at the National Research Center for Infectious Disease Diagnostic Reagents and Vaccine Engineering Technology, and asked him about the problems we encountered in designing the surface display system of E. coli and the CRISPR/Cas9 system.
8.3-8.10
- BBa_K4195164, BBa_K4195165, BBa_K4195166 were successfully characterized.
8.5
- Prepare to purify OMVs using ultracentrifugation. Getting prepared for doing experiment two o’clock in the morning.
8.8
- [Summer the girl]: I've created the greatest work of art on my NC membrane while the f**king dot-blot SHOWED ME NOTHING!!!
8.10
- The negative detection circuits (yes without input) reports fluorescence. That's the OPPOSITE results of what was expected.
8.14
- Thanks to South Putuo Temple, for the first time, the results are what I expected. The circuits with input reports STRONGER fluorescence than the circuits without input. Don’t forget to redeem tomorrow.
8.11-8.15
- BBa_K4195120 was successfully characterized .
8.15
- OMVs under TEM! We finally made it [CRYOUT]!
8.16-8.24
- BBa_K4195121 was successfully characterized .
8.24
- Pumpkin boy said he was too tired to continue the RNA EXTRACTION after getting nothing from the qPCR for several times.
8.31
- Discard the method of quantifying OMV by lipid using DiI dye. Embrace the BCA kit.
September
- interview with Prof. Yun Tian
We visited Professor Yun Tian at the School of Life Sciences, Xiamen University, and asked her about the problems we encountered in the detection of pathogenic bacteria and the induction of engineered bacteria.
9.4
- We went to Xiamen Science and Technology Museum for our mass education. Meeting those children with their innocent smile really enlightened my day. I’m energetic again for those shit results (or maybe not).
9.17
- Successfully prove that OMV can envelop plasmid.
9.18
- I’m so exhausted with these fickle circuits. A circuit with the opposite result will still have the opposite result, and a circuit with the right result will not be reversed.
