Summary

To implement MonChassis, we used several standard protocols from the metabolic engineering toolbox. We chose the RFC1000 modular cloning system (MoClo) as our primary cloning strategy. After establishing a total of 29 level 0 constructs, we assembled 49 different level 1 and 15 level 2 vectors. You can find comprehensive information regarding the underlying cloning scheme on our results page. Furthermore, we used protocols for protein modification and purification and established a modular in vitro set-up to manufacture monoterpenoids in our 3D-printed BioReactor for Enzymatic Electrosynthesis (BREES).

The materials and methods file provides detailed step-by-step guides to enable an easy reproduction of our experiments. Moreover, you find an overview of abbreviations for our new constructs and strains, as they are mentioned on our wiki pages.
