Our Approach

Our first team meeting took place on April 15th where we discussed the logistics and overview of the iGEM competition to allow potential members to gauge their interest. Meetings for brainstorming and more detailed project design occurred beginning on the 13th of May and continued into early June. The first wet lab experiments began on July 20th.

Due to shipping delays, a few critical materials were not delivered in time for the wiki freeze. Therefore, full development and characterization of our engineered phage system will be completed after the iGEM season. However we did successfully design, clone, transform, and perform phage assays with our phage tail fiber constructs.

Outreach events (collaboration, human practices, education) were either planned or fully implemented throughout the summer, with the first even being a presentation at Camp Hughes Taylor Birch on July 9th.

All work documented on this wiki was completed by us unless stated otherwise.

Team Attributions

Administration

The student team leaders were Nick Price and Daria Perminova. Nick Price and Daria Perminova were responsible for team recruitment. Daria Perminova was in charge of task delegation, maintaining communication among the team, and establishing a timeframe for task completion. Ken Nguyen, Nick Price, Daria Perminova all assisted with recruiting a principal investigator and locating lab space.

Project Design

Nick Price, Daria Perminova, and Noura Sayed contributed to literature searching and brainstorming for potential project ideas with assistance from Kevin Yehl. Nick Price significantly contributing to finalizing the design. James Mueller did literature searches for antibiotic resistance and virulence gene targets for CRISPR-Cas13.

Wet Lab

Nick Price designed plasmids and primers with assistance from Kevin Yehl. Nick Price and Noura Sayed performed PCR for plasmid components, Gibson Assembly, cloning, and transformation experiments. Nick Price and Noura Sayed carried out phage assays. Ken Nguyen assisted with initial gel electrophoresis and plasmid extraction.

Dry Lab

Alex Perminov performed AlphaFold2 modeling.

Education

Camp Hugh Taylor Birch:

Nick Price coordinated the event. Nick Price, Daria Perminova, Noura Sayed, Sam Mezzapelle designed the presentation. Nick Price, Noura Sayed, Daria Perminova presented. Daria Perminova designed the activities and collected all materials for their Implementation. Slides and microscopes were borrowed from the Hefner Museum at Miami University.

Great Lakes Science Center:

Andrew Denman coordinated the event. Andrew Denman and Sam Mezzapelle designed the presentation. Andrew Denman, Sam Mezzapelle, Daria Perminova presented. Daria Perminova planned the activities and collected most materials with assistance from Andrew Denman. Microscopes were borrowed from the Hefner Museum at Miami University. Slides were supplied by the Miami University Microbiology Department.

Miami University- Hamilton Water Festival:

Daria Perminova planned and coordinated the event with assistance from Julia Robinson, the museum educator and senior project manager at the Hefner Museum at Miami University. Daria Perminova designed the poster with assistance from Mackenzie Britton, Noura Sayed, Andrew Denman, and Sam Mezzapelle. Daria Perminova designed the activities and collected materials for their implementation. Daria Perminova designed the education bookmarks. Daria Perminova, Mackenzie Britton, Sam Mezzapelle, Andrew Denman, Noura Sayed presented.

Human Practices

CVS Pharmacy:

Noura Sayed coordinated the event, designed the presentation, and led the interview.

Dayton Children's Hospital:

Mackenzie Britton coordinated the event.

Promotional Video

Ryan Flemming edited the script and filmed and edited the promotional video. Daria Perminova wrote the script for the promotional video. Nick Price, Noura Sayed, Max Brenner, Daria Perminova served as subjects in the promotional video.

Fundraising

Daria Perminova, Nick Price, and Sam Mezzapelle researched synthetic biology companies and constructed emails for funding requests. Sam Mezzapelle wrote the submission for the Cause Exploration Grant from Open Philanthropy with assistance from Daria Perminova and Mackenzie Britton. Sam Mezzapelle wrote the submission for the iGEM Team Impact Grant.

Safety Form

Mackenzie Britton filled out the preliminary and final safety form with assistance from and editing by Nick Price, Daria Perminova, Kevin Yehl.

Collaboration

Andrew Denman coordinated meetings and led communications between the MiamiU iGEM team and the TU_Dresden iGEM team.

Alex Perminov produced protein models for the TU_Dresden iGEM team.

Other Deliverables

Daria Perminova wrote the Title and Abstract with assistance from Kevin Yehl Daria completed the Judging Form.

Recognitions

We would like to recognize Patrick Needham for his support and guidance during the wet lab experiments and associated wiki writing and the Yehl Lab for generously allowing us to use their lab space and materials, including relevant bacterial and bacteriophage strains. We would further like to recognize Bukunmi Araba who, while not an official iGEM member, volunteered to voice our promotional video.

Our Resources

Miami University irregularly offers synthetic biology as the subject focus for an advanced Topics in Microbiology course. Due to the variable nature of this course, its materials are not available online. Recently, the course focused on synthetic biology when taught by last year's team principal investigator and current secondary principal investigator, Dr. Xin Wang. Five of our members have had the opportunity to take this course in a current or previous semester. Daria Perminova, Ken Nguyen, and Max Brenner took the course in the spring of 2021 and Nick Price took the course in the spring of 2020. One of our members, Andrew Denman, is currently enrolled.
