Many of the parts registered by our team this year can be cateogrised into 3 part collections:

Throughout the duration of our project this year, our dry lab team was able to create over 20 peptides against effector protein in Puccinia striiformis. By evaluation of each peptides’ modelled efficacy and confidence score, 15 inhibitory peptides against 2 different effector proteins were added to the Registry of Standard Biological Parts. A comprehensive review of literature highlighted various effector proteins associated with the pathogenic mechanisms of stem rust fungi. Several of these proteins were selected for use in our project this year and added to the Registry of Standard Biological Parts.

With our team deciding to use FRET as an indicator of protein binding in our wet lab experiments, the fluorescent protein mCerulean was added to each of the fungal and plant proteins at either the N or C-terminal. Modelling was performed using AlphaFold to detect disruptions to the original proteins’ structure following the addition of the fluorescent tag. Successful candidates were synthesised in the lab and added to the Registry of Standard Biological Parts.
