Establishment of partnerships:
At the iGEM summer party online meeting on July 16, 2022, we met the SZPT-CHINA team. Through communication, we learned that their project GSH production module needs to control the output of GSH to reach the ideal data; our project is based on YeastFab standardized DNA assembly platform to quickly find the optimal enzyme activation ratio. The similarity between the two projects is an important reason why we decided to establish cooperative relationship.

SZPT-CHINA→NEFU_China:
The SZPT_CHINA team invited us to participate in the online scientific popularization lecture on "Walking into Synthetic Biology" on August 6, with an audience of more than 300 non-biological college students. And we gave a lecture on the theme of "synthetic biology is not a profound science" to let more non-biological specialties. Students understand that synthetic biology is closely related to our daily life and stimulate their enthusiasm to actively learn about synthetic biology.

Our team encountered problems in the experiment and did not find out reason for a long time. On August 9, we asked the SZPT-CHINA team for help. They visited Wenwen Xiao and Shengwei Fu, two experts from the Quantitative Center of the Synthesis Institute of Shenzhen Advanced Technology Research Institute . We benefited a lot , solved the problem and made improvement.

NEFU_China: We have used PCR to evaluate the results for a long time, but found that the results of this proof are unstable. Do you have any good methods to solve it? Wenwen Xiao & Shengwei Fu: It seems better to extract the genome for evaluation. NEFU_China: Teacher, we lack experience in genome PCR. Is there any experience to learn from? Wenwen Xiao & Shengwei Fu: There are many uncertainties about genome PCR, but there are also some ways to maximize the efficiency of genome PCR, such as controlling the length of PCR fragments at 200 to 2000bp. |
NEFU_China⇋SZPT_CHINA:
At the online exchange meeting on July 17, we had in-depth exchange meeting with the SZPT-CHINA team and discussed each other's projects, and put forward our views on the SZPT-CHINA team's project, such as: Is bacillus a safe strain? Does visible light interfere with the optical control system? In order to analyze the output of target production, we share the method of selecting the high-yielding antioxidant strains by using hydrogen peroxide stress. The suggestion we proposed greatly contributed to SZPT-CHINA team in experimental progress.

In terms of modeling, the SIPT-CHINA team found that the GSH output of the modified Acetella is too low. Our team suggested that the three factors affecting the production of GSH be found through the response surface, and the corresponding optimization conditions are found to build an output prediction model. They accepted our suggestion and achieved the expected results in the later experiment.
SZPT-CHINA was inspired by our three-line poetry activity on synthetic biology. So we cooperated in three microbial hymn activities, using the WeChat official account as a platform to promote synthetic biology, in the form of poetry, so that students who are not major in biology can get access to synthetic biology.
We NEFU_China members actively helped SZPT-China to promote their project, and we had in-depth conversation .
