Section 1 : Introduction

Biology is different from other engineering fields due to a lack of modular nature of the structure and function, which means the expression of gene will change when transferred from one species to another. What synthetic biology is dedicated to is the modularization and standardization of functional gene, to make the expression of it predictable in different chassis organism. Our project has registered 8 basic parts and 6 composite parts, all qualified for standardized requirement of RFC[10]. We have also tested these parts. We hope that everyone can utilize our parts based on our research through registry and that the information for parts can further be completed so that more people can make use of them.

Section 3 : Improvement

Fusion Protein (FP) and LanM(Lanmodulin) are proteins binding Lanthanide that can be used to purify the water polluted by REE mining industry. This basic part has been registered by iGEM2017_HUST as BBa_K2336033. We improved its function and registered as new parts BBa_K4468007 and BBa_K4468006.
Firstly, we optimized the codon of this gene according to the codon preference to improve its expression level.
Secondly, research shows that LanM is a strong Lanthanide binding protein with three strong binding site. we construct a LanM-dLBT fusion proteins named FP and expressed it, hoping that they could show a better binding activity. We also want to recycle the engineered strain for the sake of safety and economy concern. So we bind the Si-tag with the protein to bind the cell to SiO2 in nature sand.
Then, we predicted the 3D structure of FP. The docking result shows that the Tb3+ have an affinity to the EF-hand domains of the fusion protein.
Therefore, we started to build the regulation system and achieved success in expression.
