Our team collaborated with the team HKU this year. Since we are focusing on the expression and purification of 3-hydroxybutyrate dehydrogenase (CF3HBD) and lactam-binding protein (NitR) this year, our team has been considering different protein expressing plasmids. This year team HKU offered us the pET28a plasmid, which is used for in the previous literature. This plasmid is used for the expression of 3-hydroxybutyrate dehydrogenase (CF3HBD) and its 6xHis tag allowed us to do efficient purification.
To further enhance the production of the two interested plasmids, the HKU team also offered us a modified version of pET28a (pMET28a). Although due to time constraints, our team did not have time to test whether the use of pMET28a would increase the yield of the proteins in our system.
