Heavy chain human ferritin | BBa_K1189025 | To recycle our enzyme-expressing microbes and cellulosome complexes after the degradation of fabric, magnetism is the key point. Utilizing the human ferritin heavy chain provided by iGEM13_Calgary, we are able to generate magnetoferritin via the chemical modification of the original design - the addition of ammonium iron (II) sulfate into the medium. |
eforRed, red chromoprotein | BBa_K592012 | Proving the functionality of our giant cellulosome complex is indispensable for our project. The eforRED chromoprotein provided by Uppsala-Sweden is the best candidate for our reporter protein. We developed 3 different fusion variants of eforRED, each testing for a specific molecular interaction in our construct. The bright color and high expression level of eforRED facilitate our observation and data collection to a great extent. |
PETase-5, PET hydrolase | BBa_K3715005 | The degradation of polyester fiber is a crucial part of the project FabRevivo. Due to the low catalytic efficiency and thermostability of the wild-type PETase, we craved a highly efficient PETase mutant that can degrade high-crystallinity polyester fiber under high temperatures. Thanks to the Super-5 PETase mutant provided by TJUSLS_China, the degradation of this recalcitrant artificial fiber will no longer be a time-consuming process. Furthermore, we developed a Dockerin-fused variant of this miraculous PETase mutant. |
