This year, the 2022 Edinburgh-UHAS_Ghana iGEM Team decided to make a collection of parts centred around two main themes: Cell Free Systems and JUMP assembly. Across all facets of our project, the final goal is always a cell free device; the biosensors, the hydrogel bioremediation devices and the Silica-immobilised PETases. This collection of parts provides an excellent framework for any teams looking to immobilise enzymes and produce fluorescent aptamers. Furthermore, JUMP assembly (a type IIS assembly method Accepted by iGEM) links together all our parts, as this was the only assembly technique we used for DNA assembly. JUMP combines the modularity of BioBricks and PhytoBricks with the convenience of Golden Gate or MoClo assembly. Having designed all our parts to be JUMP compatible, this part collection also serves as a valuable toolkit for teams looking to use JUMP assembly. We have added many common parts required for synthetic biology, as well as designed new utility parts (See BBa_K4390006-8) to improve modularity even further. This meant that from a relatively small number of basic parts, we were able to assemble a large combinatorial array of composite parts. We hope that future teams will utilize parts in our collection to continue excellent JUMP-assembled synthetic biology work.

We chose BBa_K4390006 as our best part, the JUMP N-adapter. We used this part extensively in modularisation, as it allowed us to exclude type N parts from assembly, when we wanted to form transcriptional units using O and C parts we had available. This was especially useful for any tagging, as it meant we could quickly and easily produce untagged parts without having to order in new parts. This part is compatible for all JUMP assemblies, and can be used in any constructs.
