Application of photoinducible promoter in the biosynthesis of patchoulol in yeast

We used yeast to synthesize patchoulol. There are several key enzyme genes in the patchoulol synthesis pathway, and these elements can be optimized in yeast through gene replacement, amplification of gene elements, gene fusion expression, gene cluster integration and combination with resistant elements. Increasing the supply of the precursor substrate FPP somewhat increases its total flow to patchoulol and promotes the synthesis of patchoulol.

Photoinducible Promoter

Light-regulated promoter (a region of DNA where transcription of a gene is initiated) that are specifically responsive to different wavelengths allows spatiotemporal control of gene expression.

Patchoulol Synthase

Patchoulol synthase (EC 4.2.3.70) is an enzyme with systematic name (2E,6E)-farnesyl-diphosphate diphosphate-lyase (patchoulol-forming). This enzyme catalyses the following chemical reaction.

What is Pogostemon cablin?

Pogostemon cablin, belongs to perennial aromatic herbs or semi-shrubs, is one of the first “China Lingnan authentic medicinal materials protected varieties", and one of the famous "top ten southern China medicines". It is mainly planted in Indonesia, Singapore, Vietnam, Malaysia, China and other regions.

Curative effect of Pogostemon cablin?

As "Herbal Justice" (An important classic books of Chinese Medicine) said: 藿香清芬微温,善理中焦湿浊痰涎,为醒脾快胃,振动清阳妙品,所以适用于湿阻中焦证. Patchouli is used in many prescriptions, such as Huoxiang Zhengqi powder and Lotus Qingwen capsules.

What is Patchouli oil?

Patchouli oil is processed by distillation on the dry ground part, and its main components are heavy cyclic aromatic hydrocarbons, ketones, alcohols and terpenoids, as well as some alkaloids. Patchouli oil is equipped with 17 pharmacological effects, such as antibacterial, anti-inflammatory, anti-pain, anti-depression, anti-platelet, anti-gastric ulcer and anti-photoaging, which can be used as medicine; the other is its unique fragrance and significant aroma function, which can be used as a spice and food additive. Patchouli oil is widely used in daily chemical products, drugs and food industries, and has a broad market prospect.

What is Patchouli?

1. Patchouli is a sesquiterpene, one of the most abundant components in patchouli oil.
2. Patchouli is an important indicator to evaluate the quality of patchouli and its essential oil in Chinese Pharmacopoeia, and is also considered as the main contributing component to the pharmacological efficacy and aroma of patchouli oil.
3. It has reported a variety of promising medicinal activities, including anti-influenza virus, antidepressant, anti-vascular relaxation, lung protection, brain protection, anti-ulcer, anti-colitis, prebiotics, anti-inflammatory, anti-cancer, and protective activity against metabolic diseases.

Application and productivity of Patchouli

1 Patchouli has reported a variety of promising medicinal activities, including anti-influenza virus, antidepressant, anti-vascular relaxation, lung protection, brain protection, anti-ulcer, anti-colitis, prebiotics, anti-inflammatory, anti-cancer, and protective activity against metabolic diseases.
2. At present, the global annual production of Patchouli and its mixed essential oil is about 1,300t, but the annual demand can reach 2,000t, which exceeds short supply. Indonesia supplies 90% of the production, while China produces less than 10%, leaving ample room for growth.)

At present, there are three kinds of methods to synthesize Patchouli, namely plant extraction, chemical synthesis and microbial fermentation synthesis:
(1) Among them, plant extraction method is the main method of producing patchouli alcohol, but this method requires a large number of patchouli plants as raw material to occupy agricultural farmland; the yield and quality of patchouli alcohol are greatly affected by geographical location, climate environment and plant quality; moreover, the process of extracting patchouli alcohol is extremely energy consuming, which is not conducive to environmental protection.
(2) Although the chemical synthesis method can synthesize patchouli alcohol with high purity, it has many steps, complex operation, high synthesis cost and very low yield, which cannot be applied to large-scale industrial production.
(3) The use of microbial fermentation to synthesize patchouli alcohol has the advantages of low cost, conducive to environmental protection and create economic benefits. At present, most S. cerevisiae or Corynebacterium glutamate are used as the host bacteria.
